OK, let's set up the call for the other topics (a and b above).
We wouldn't just do his bioinformatic analyses and not the rest. le, the bioinfo
platform will cost the same for 1 person vs 100 or more once the system is
purchased. But I'll look into other funding for this.
Obviously (and hopefully this was clear), Jeffrey's enrollment in the study has absolutely
no bearing on whether he helps fund the bioinfo or not. I wouldn't do that to him.
He's in regardless. As we look to fund this piece, it just seemed worth asking.

Thanks - this is research so nothing can be 100% certain ,but, to my
knowledge, our study will have greater NGS data privacy than any
other similar medical genomics research effort.
Afterwards, if Jeffrey still wanted to share his data through the PGP
or other efforts, we could help facilitate that.

Sorry for the delayed response. We just got word from
the IRB and we're finally ready to push forward with
the YPO study.
Lesley, I wanted to discuss a couple things with Jeffrey by phone at his
convenience including:
a) getting a whole genome through Illumina puts him a little bit outside of the
workflow pipeline for the YPO project. Ie, those
participants are paying $1000 each and will be getting
their exomes sequenced through Ambry, a thumbnail
drive of their compressed genomic data, and an
annotated report of 56 ACMG gene hits. I think it's
worth getting him into this queue as well so he goes
through the same pipeline as the YPO folks and others
in our study. Last time I discussed with him he
definitely wanted a whole genomes through Illumina
for 411,000 (which is still fine) but would he be
interested in also getting the SI000 exome to also go
through the normal pipeline? Although the exome data
should be redundant with the whole genome data, I
think it's worth it for the thumbnail drive and the
ACMG gene summary. We'll also be able to check for
concordance between the 2 datasets.
b) Jeffrey has samples (including fibroblast cell culture from his skin biopsy)
at HMS. Do I have his permission to request these

samples from the PGP for inclusion in the YPO study?
(The samples can be used for both studies if he
wishes.) Our YPO study may need a self-collected
saliva sample for which I can mail him a kit. So, I
wanted to discuss these sample issues with him as
well.
c) Lastly, we actually still need some bioinformatic and genetic counselor
support for the YPO study - not just for his sample but
all of our 80 plus YPO study subjects (from the Austin
chapter). As I mentioned, all 80+ have already paid
$1000 each for the sequencing but we still need to
cover bioinformatics and a part-time GC for all
particpants. In the past, months ago now do to the
IRB delay, Jeffrey was generous enough to offer to
pay for sequencing of YPO members. Since that's
already covered, I'm wondering if he would consider
donating financial support for bioinformatic
infrastructure for this study.
